Minkie Spiro absolvierte ihr Studium an der Central St. Martins School in London mit einem First-Class-Honours-Abschluss und machte ihren Master in Filmregie am Royal College of Art in London. Ihre frühen Kurzfilme wurden auf mehreren internationalen Filmfestivals gezeigt und ausgezeichnet. Spiro feierte ihre größten Erfolge mit TV-Serien, unter anderem mit Call the Midwife und Downtown Abbey. 2004 wurde sie für einen BAFTA als beste Nachwuchsleistung nominiert.
